PGK1T terminator

  • The PGK1 terminator (PGK1T) is a downstream regulatory sequence of the PGK1 gene in Saccharomyces cerevisiae, responsible for terminating the transcription of this gene. The PGK1 gene encodes phosphoglycerate kinase, an essential enzyme in glycolysis that catalyzes the conversion of 1,3-bisphosphoglycerate to 3-phosphoglycerate, producing ATP in the process. The PGK1 terminator ensures precise termination of transcription, preventing read-through and ensuring proper 3' end processing and stability of the mRNA.In synthetic biology and metabolic engineering, PGK1T is widely used as a reliable terminator, as it provides efficient and stable termination signals under various conditions. It is often paired with strong promoters in yeast expression vectors to ensure accurate transcription and translation termination of foreign genes, contributing to higher expression levels of the target protein.
